枇杷脫水素基因EjDHN5抗低溫脅迫功能研究

【摘要】:為研究枇杷(Eriobotrya japonica Lindl.)脫水素基因(Ej DHN5)在枇杷抗低溫脅迫中的功能,將其在煙草中進行過量表達,其中表達水平比較高的兩個株系L24和L26被用來進行功能研究。野生型和轉(zhuǎn)基因煙草苗分別用0、2、4和8μmol·L~(-1)的甲基紫精(Methyl viologen,自由基發(fā)生劑)處理并進行光照培養(yǎng),結(jié)果顯示,4和8μmol·L~(-1)甲基紫精處理使野生型煙草苗存活率分別下降到73.6%和56.9%,光系統(tǒng)Ⅱ(PSⅡ)活性顯著降低,而轉(zhuǎn)基因苗的存活率仍維持在100%,PSⅡ活性明顯高于野生型。對六葉期煙草的葉盤進行甲基紫精處理,野生型煙草葉盤的葉綠素含量顯著低于轉(zhuǎn)基因株系,而活性氧含量和MDA含量明顯高于轉(zhuǎn)基因株系。對野生型和轉(zhuǎn)基因煙草苗進行低溫處理,轉(zhuǎn)基因株系生長狀況明顯優(yōu)于野生型,活性氧含量低于野生型,膜質(zhì)過氧化程度輕于野生型。這些結(jié)果表明,過量表達EjDHN5能提高煙草的抗氧化脅迫和抗低溫脅迫能力,推測EjDHN5在提高枇杷抗低溫能力方面起著重要作用可能與其能提高抗氧化脅迫能力有關。
