Panx1基因過表達對人肝癌Hep3B細胞增殖及遷移的影響

【摘要】：目的探討Panx1基因過表達對人肝癌Hep3B細胞增殖及遷移的影響。方法構建Panx1過表達慢病毒質(zhì)粒和過表達空載慢病毒質(zhì)粒,包裝生產(chǎn)慢病毒,分別轉(zhuǎn)染Hep3B細胞(過表達組、對照組)。用Western blotting法檢測Hep3B細胞Panx1、上皮鈣黏蛋白(E-cadherin)及波形蛋白(Vimentin)表達,CCK-8法檢測細胞增殖能力(OD值),劃痕實驗檢測細胞遷移能力(相對遷移距離、劃痕愈合率)。結果過表達組Panx1、E-cadherin蛋白相對表達量較對照組高(P均0.05),Vimentin蛋白相對表達量較對照組低(P0.05);各時間點(0 h除外)OD值較對照組低(P均0.05);相對遷移距離較對照組低,劃痕愈合率較對照組高(P均0.05)。結論 Panx1基因過表達可抑制Hep3B細胞增殖與遷移。
[Abstract]:Objective to investigate the effect of overexpression of Panx1 gene on proliferation and migration of human hepatoma Hep3B cells. Methods Panx1 overexpression lentivirus plasmids and empty lentivirus plasmids were constructed and packaged to produce lentivirus. The lentivirus was transfected into Hep3B cells (overexpression group, control group). The expression of Panx1, epithelial cadherin (E-cadherin) and vimentin (Vimentin) was detected by Western blotting assay, cell proliferation ability (OD value) was detected by CCK-8 assay, and migration ability (relative migration distance, scratch healing rate) was detected by scratch test. Results the relative expression of Panx1,E-cadherin protein in the overexpression group was higher than that in the control group (P < 0. 05), the OD value at each time point (except 0 h) was lower than that in the control group (P 0. 05), and the relative expression of), Vimentin protein in the overexpression group was lower than that in the control group (P 0. 05). The relative migration distance was lower than that of control group, and the rate of scratch healing was higher than that of control group (P 0.05). Conclusion overexpression of Panx1 gene can inhibit the proliferation and migration of Hep3B cells.
