【摘要】：[目的]觀察姜黃素對(duì)SW620細(xì)胞及裸鼠NKD2、CXCR4表達(dá)水平的影響。[方法]通過MTS檢測(cè)姜黃素對(duì)SW620細(xì)胞活性的影響;運(yùn)用Western blot、Real-time q PCR檢測(cè)不同濃度姜黃素對(duì)SW620細(xì)胞及轉(zhuǎn)染NKD2 si RNA后SW620細(xì)胞內(nèi)NKD2、CXCR4蛋白和m RNA表達(dá)的影響。雄性BALB/c裸鼠8只,隨機(jī)分成對(duì)照組與姜黃素組,每組4只。采用皮下接種SW620細(xì)胞。造模后第7d開始給對(duì)照組與姜黃素組分別注射對(duì)照液和100mg/kg·d濃度的姜黃素懸液,連續(xù)用藥14d。給藥后檢測(cè)瘤體體積,通過Real-time q PCR檢測(cè)瘤體組織內(nèi)NKD2及CXCR4表達(dá)水平。[結(jié)果 ]姜黃素能明顯抑制SW620細(xì)胞的增殖活性,顯著性上調(diào)SW620細(xì)胞內(nèi)NKD2表達(dá)及下調(diào)CXCR4的表達(dá),且呈劑量依賴性。轉(zhuǎn)染后發(fā)現(xiàn)能上調(diào)姜黃素處理后的腫瘤細(xì)胞CXCR4的表達(dá)水平。體內(nèi)實(shí)驗(yàn)表明姜黃素抑瘤率為30.69%。通過q PCR發(fā)現(xiàn)瘤體組織中NKD2的表達(dá)量增加,CXCR4的表達(dá)量顯著性降低。[結(jié)論]姜黃素均能從體內(nèi)外明顯抑制結(jié)腸癌細(xì)胞SW620的生長(zhǎng),其作用機(jī)制可能通過上調(diào)抑制基因NKD2的表達(dá),抑制Wnt信號(hào)通路,進(jìn)而下調(diào)腫瘤細(xì)胞CXCR4表達(dá),從而起到抑制腫瘤侵襲轉(zhuǎn)移的作用。
[Abstract]:[objective] to observe the effect of curcumin on the expression of NKD2,CXCR4 in SW620 cells and nude mice. [methods] the effect of curcumin on the activity of SW620 cells was detected by MTS, and the expression of NKD2,CXCR4 protein and m RNA in SW620 cells and SW620 cells after NKD2 si RNA transfection was detected by Western blot,Real-time Q PCR. Eight male BALB/c nude mice were randomly divided into control group and curcumin group with 4 rats in each group. SW620 cells were inoculated subcutaneously. The control group and curcumin group were injected curcumin suspension with control solution and 100mg/kg d concentration respectively on the 7th day after modeling. The volume of tumor was measured after administration, and the expression of NKD2 and CXCR4 in tumor tissue was detected by Real-time Q PCR. [results] curcumin significantly inhibited the proliferation of SW620 cells, significantly up-regulated the expression of NKD2 and down-regulated the expression of CXCR4 in SW620 cells in a dose-dependent manner. After transfection, it was found that curcumin could up-regulate the expression of CXCR4 in tumor cells treated with curcumin. The inhibition rate of curcumin was 30.69% in vivo. Q PCR showed that the expression of NKD2 increased and the expression of CXCR4 decreased significantly. [conclusion] curcumin can obviously inhibit the growth of SW620 in colon cancer cells in vitro and in vivo. The mechanism of curcumin can inhibit the expression of Wnt signal pathway by up-regulating the expression of NKD2 gene, and then down-regulate the expression of CXCR4 in tumor cells. Thus it can inhibit the invasion and metastasis of tumor.
