　　 組培、嫁接快繁體系的建立被廣泛應(yīng)用于現(xiàn)代葡萄栽培生產(chǎn)中,以減少生物和非生物脅迫造成的危害。為葡萄種植產(chǎn)業(yè)提供無(wú)病毒砧木是控制病害的有效途徑。本研究中,以"SO4"、"101-14"、"5BB"、"110R"和"1103P"五種常見(jiàn)的葡萄砧木品種為外植體,通過(guò)建立組培快繁體系快速獲得無(wú)病毒葡萄砧木�？旆斌w系建立結(jié)果顯示,快繁體系中適合不同砧木啟動(dòng)培養(yǎng)的培養(yǎng)基為MS基本培養(yǎng)基添加0.2 mg/L IBA、1.0 mg/L 6-BA、0.5 mg/L KT、4.0 mg/L腺嘌呤;適合不同砧木繼代的培養(yǎng)基為WPM基本培養(yǎng)基添加0.2mg/L IBA,且增殖系數(shù)在1.6～4.4之間。病毒檢測(cè)結(jié)果顯示,單次RT-PCR檢測(cè)三種病毒比雙重和三重RT-PCR更有效。通過(guò)不斷繼代獲得無(wú)GLRaV-3,GFKV和GRSPa病毒組培苗進(jìn)行大棚移栽馴化。幼苗不同霍格蘭溶液中室溫自然光照下馴化2周,移栽到溫室中混合培養(yǎng)基質(zhì)中,縮短馴化周期。該方案適用于5個(gè)葡萄藤砧木品種的快速繁殖,可用于無(wú)病毒葡萄砧木的商業(yè)化生產(chǎn)。
