過(guò)表達(dá)篩選發(fā)現(xiàn)果蠅miR-2參與調(diào)控線粒體穩(wěn)態(tài)(英文)

【目的】真核細(xì)胞內(nèi)線粒體穩(wěn)態(tài)由多條分子通路共同調(diào)控,其中miRNA的重要性在最近日益凸顯。但是對(duì)miRNA如何調(diào)控線粒體穩(wěn)態(tài)仍缺乏全面準(zhǔn)確的認(rèn)識(shí)。本研究旨在探究miRNA對(duì)線粒體穩(wěn)態(tài)的調(diào)控作用及機(jī)制。【方法】在黑腹果蠅Drosophila melanogaster幼蟲脂肪體細(xì)胞中對(duì)miRNA進(jìn)行過(guò)表達(dá)篩選,監(jiān)測(cè)線粒體形態(tài)變化。通過(guò)生物信息學(xué)方法預(yù)測(cè)參與調(diào)控線粒體穩(wěn)態(tài)的miRNA下游靶標(biāo),并通過(guò)RNAi敲低實(shí)驗(yàn)檢驗(yàn)靶標(biāo)基因?qū)粒體形態(tài)的影響!窘Y(jié)果】在黑腹果蠅幼蟲脂肪體中監(jiān)測(cè)了過(guò)表達(dá)106個(gè)miRNA對(duì)線粒體形態(tài)及果蠅發(fā)育的影響,發(fā)現(xiàn)21個(gè)miRNA過(guò)表達(dá)引起脂肪體發(fā)育異常,其中10個(gè)miRNA過(guò)表達(dá)可以導(dǎo)致幼蟲期致死,11個(gè)miRNA過(guò)表達(dá)導(dǎo)致蛹期致死。過(guò)表達(dá)miR-2導(dǎo)致黑腹果蠅脂肪體細(xì)胞中線粒體異樣聚集。序列分析表明miR-2很可能靶向調(diào)控Pink1基因。Pink1基因表達(dá)量確實(shí)因miR-2過(guò)表達(dá)而下調(diào)。遺傳互作實(shí)驗(yàn)發(fā)現(xiàn)過(guò)表達(dá)Pink1基因可以拯救過(guò)表達(dá)miR-2引起的線粒體異樣聚集。【結(jié)論】結(jié)果說(shuō)明miR-2很可能通過(guò)靶向Pink1基因參與調(diào)控線粒體穩(wěn)態(tài)。
