慢性應(yīng)激通過減少毛囊黑色素細(xì)胞和酪氨酸酶活性導(dǎo)致雌性C57小鼠毛發(fā)顏色變淺(英文)

越來越多的證據(jù)表明壓力可能會導(dǎo)致頭發(fā)顏色發(fā)生變化,但其潛在機(jī)制尚不完全清楚。本研究采用雌性C57BL/6小鼠腳底電刺激結(jié)合束縛來建立慢性應(yīng)激小鼠模型,并用比色法檢測小鼠皮膚和B16F10黑色素瘤細(xì)胞中黑色素含量和酪氨酸酶活性;通過酶聯(lián)免疫吸附實驗(ELISA)測定小鼠皮膚中腫瘤壞死因子α(tumor necrosis factorα, TNF-α)、白細(xì)胞介素1β(interleukin-1β, IL-1β)和白細(xì)胞介素6 (interleukin-6, IL-6)含量;通過免疫熒光染色評估小鼠皮膚中核因子κB (nuclear factorκB, NFκB)/p65亞基的含量。結(jié)果顯示:C57BL/6小鼠在慢性應(yīng)激下由于皮膚中的毛囊黑色素細(xì)胞和酪氨酸酶活性降低,其毛皮顏色從暗色變?yōu)樽厣。同時,慢性應(yīng)激小鼠皮膚炎癥反應(yīng)增加,表現(xiàn)為皮膚中NFκB活性和TNF-α表達(dá)增加。在體外,TNF-α以劑量依賴性方式降低B16F10黑色素瘤細(xì)胞中黑色素生成和酪氨酸酶活性。以上結(jié)果表明,慢性應(yīng)激通過降低雌性C57BL/6小鼠的毛囊黑色素細(xì)胞和酪氨酸酶活性來誘導(dǎo)皮毛顏色改變,而TNF-α可能在應(yīng)激誘導(dǎo)的...
